No, live blood analysis is not enough for this. Just because they are no longer in your bloodstream doesn't mean they aren't in the tissues, which is where they like to stay until they are ready to replicate.
Also, as arermix said, for some, the bacteria never goes away - it's about getting symptom free. Usually we say that a person needs to be symptom free for 2 months before stopping treatments. This is to get at least 2 life cycles past the stop of symptoms, to hopefully catch any remaining bacteria.
But don't forget, Lyme rarely is found by itself, so you could easily still have other infections that need to be treated.
