Thanks for the info on the magnets! so you think your dog had some kind of EMF sensitivity (creating free radicals) which the magnet relieved?
So as far as DNA string or oligos you would need to purchase them from places like IDT or invitrogen. You would need to clone the double stranded dna and there are few methods you can do that with. As far as introducing the final product plasmid into ecoli that would be through multiple ways but the most common is heat shock which works very well. I haven't worked with yeast but reading around it seems that a good method that won't need too much equipment is the (lithium acetate procedure). There may be others out there.
You want to make sure that there is a good selection marker (puromycin or others) so that only the yeast with your plasmid grows out.
Now it seems like perhaps you don't need CRISPR in order to introduce the genes into yeast, just the cloned plasmid which is enough to make your gene/protein. But i guess with CRISPR you can permanently put the gene into the yeast and not have to think about
selection with antibiotics.
