The higher magnification can help, but does not make a gigantic difference.

The main thing to remember is that you want a 100x objective.
That, paired with 10x eyepieces will give you a total magnification of 1000x.

They don't make objectives higher than 100x.
So to get a total magnification of 2500x, the eyepieces would be 25x rather than the more typical 10x.

This does not provide any additional quality. It's akin to digital zoom on a camera; it's just making the image larger for you to see, not actually creating any additional magnification of the object.
25x eyepieces shouldn't cost very much, but personally, I'd stick with 10x.

I've never used phase contrast, but the images do look really good.
Very expensive though.

Are they ( spiros/ coinfections) in our saliva?

Oral spiros and Lyme spirios may not dwell in same places. Or they may ... I will look for evidence of Lyme spiros in saliva. So gross. So interesting. So sad.

I'm seriously considering getting a scope. They aren't cheap so if I spend the money I don't want to have to do it twice.

Spirochetes are a normal part of the oral flora. Not sure how you would differentiate them from lyme spirochetes. They're pretty crazy to watch none the less. Saliva is pretty cool to look at in general actually. Sometimes i'll add a drop of blood and those little critters freak out and swarm

Salty i found that 20x eyepiece let in a lot less light .. the way i image is with a .5 extesion tube and a cam with a direct seriel port to a 21 in computor screen ...the frame rate is great ..resolution ok but really save the ol peepers ..i am working on converting to darkfield with polarizing film and led light source ...i went used at first to learn and i knew the imaging was going to be a lil investment ..scope 150 ..cam 150 slides stain etc 50. Darkfield expermient is costing 30$..may get a new 100x oil lens too ..still

blue, I didn't know you got a scope! That's great! I don't know what your darkfield experiment involves, but you can make a darkfield annulus with permanent marker and some plastic sheeting. You can also buy darkfield inserts from Ebay for about the price you mentioned, so maybe that is what you are doing.

I agree about the 20x oculars. The amount of light let through is greatly diminished compared to 10x oculars. I have found 15x oculars work ok though, and is a good compromise. The microscope camera is a good suggestion but definitely runs into extra money.....

It gives amazing results, and each stain takes about 30 seconds.

I am fascinated by all this and hope more people comment. Are those scientists claiming because they couldn't culture it, it's not a microbe or it isn't there??? Sure sounds like it.
