One thing I have noticed is that borrelia does not immediately die from valacyclovir, but dies some days later, and the die off is accelerated if borrelia is later stressed with something else like desloratadine or common antibiotics like minocycline and azithromycin.
The way valacyclovir normally works when treating herpes is that it is first metabolized to acyclovir. Acyclovir is relatively non-toxic to normal cells, but in infected cells the viral thymidine kinase enzyme phosphorylates acyclovir, and this metabolite blocks the DNA polymerase.
The problem seems to be that borrelia phosphorylates acyclovir much more readily than a small number of herpesvirus-infected cells, forming large amounts of this metabolite which becomes toxic to human cells, causing severe side effects even at low doses.
Since the phosphorylated acyclovir gets released during die off and herx, it seems to help to not let too much of this build up, and wait for the herx to finish before taking more valtrex.
Who knew the MOA/Pharmocokinetics of Various Drugs used to treat: Malaria, Herpes, Leprosy (yes Leprosy!), and last but certainly not least, TBI's (tick borne infections like Borreliae, Babesiae, and Bartonellae!). the 3 BBBs....!!!

I had been taking Claritin (Loratidine) and faced a days-long Herx...also Lysine does something similar "Lyse"'s or cuts/dissolves/splits
open HSVs. Like 70-80% or so of the adult population, I also carry an HSV and get painful nerve pain whenever the Bbb's and Cos rear their ugly heads... stress/load increases...base of spine, neck area, full body pain, post exertional malaise..for days!... So now I know the MOA or pharmocokinetics of Acyclovir slightly better thanks to your great explanation about

I am leery of taking any of the following because of severe side effects: Disulfiram (neuropathy), Dapsone (bc Leprosy drugs are heavy), Claritin (bc Manganese channels effectively kill Bbs n Co days later for prolonged Herx with psychiatric manifestations!)...I have a similar theory about
cannabis Abx Fx, it kills active infections and these toxic metabolites cause paranoia/psychosis something about
the D1 channel I'm out of my depth somewhat here....but Minocycline and Z-Pak (Azithromycin) make me nervous for my mental health as I have no safe way of going about
it whenever I can get them prescribed readily, the doctor/clinics dismiss my Dx/Sx entirely resulting in PTSD! Now that Ketamine is approved for the treatment of depression, maybe I won't have to suffer as much next time? Anyway...

I would say taking Valtrex helps to reduce your overall viral load and that's a good thing. If you can tolerate the Side Fx through the healing just take it slow and practice self-care/ sleep hygiene until the healing is over then repeat if desired or taper down until it is manageable...
And about
the Manganese channels' disruption for further readings, not so off-topic bc this is very complex organic chemistry here when it comes down to it!
