"Pfizer Inc. (NYSE: PFE) announced positive topline results from the final overall survival (OS) analysis of the TALAPRO-2 study. The study evaluated TALZENNA® (talazoparib), an oral PARP inhibitor, in combination with XTANDI® (enzalutamide), an androgen receptor pathway inhibitor, in patients with metastatic castration-resistant prostate cancer (mCRPC).

Results showed a statistically significant and clinically meaningful improvement in OS for all patients and those with homologous recombination repair (HRR) gene-mutated mCRPC, compared to XTANDI alone. This makes TALZENNA plus XTANDI the first and only PARP inhibitor-ARPI combination to significantly improve survival in mCRPC patients, regardless of mutation status.

The safety profile was consistent with known profiles of each medicine. Detailed results will be presented at an upcoming medical congress and shared with global health authorities for potential label updates."

Great to hear! And, as usual, if it works for the CR guys, it will probably work really well for the CS guys. It’s too bad that we’ll probably have to wait 5 years for another study to prove it. Aren’t we to the point where things can be streamlined after the CR guys are taken care of?

@Mattam, to your comment about streamlining trials, we are getting there. Slowly, very slowly, but we are making progress towards that end.

The key sentence is: "Results showed a statistically significant and clinically meaningful improvement in OS for all patients and those with homologous recombination repair (HRR) gene-mutated mCRPC, compared to XTANDI alone." I have read too many of these where the $10K/month drug extends life by 2 months which is not the most exciting news, especially if there is a new side effect profile.

Never quite know what to make of these statements. Given the slow nature of PCa and older population, getting long term data is difficult then comes the evaluation process.

Hey Prato,
I certainly hope they can find a way to streamline these trials. I don’t know if it’s the case with other cancers (maybe BCa?), but our hormone dependent PCa may be fairly unique in that we can hold most of PCa cells in “suspended animation” for quite a long time by removing or blocking the hormone. It seems to me, that it’s becoming apparent that any agent that can kill the PCa cells that have somehow bypassed the hormone requirement would work really well on CS men. I think we need to take care of our CR brothers first, and it’s great reading about treatments that are helping advanced guys live longer. But it’s frustrating knowing that the same treatment will require years to be approved for CS guys who have limited ADT/ARPI windows.

Hey, I noticed that you’re at 9979. That’s quite a dedicated accomplishment!!! Too bad we all can’t party together like it’s 1999 when you hit 9999.

I agree. But let’s not forget that when Zytiga and Xtandi went into trials on CR men the outcomes weren’t exactly stupendous. If I remember correctly most men got an extra few months to an extra year. But, when sequenced while men are still CS, survival is being measured in years instead of months. I think the endpoints still haven’t been reached in some of the trials. I sense that Pluvicto and Pluvicto-like “drugs” could be highly successful in a CS setting.

Good catch matter. What's interesting to note, is that my guess is the first 3000 posts came in the first year I was here. Thats how out of my mind I was LOL. I remember going to one of the gatherings after the first year, and I was asked by Jerry (Redwing, great guy, not here as much anymore but does show up on occasion) how on earth I managed to put up so many posts in such a short time LOL>

I have high hopes for AI streamlining these trials and doing so much more for medicine.
